A study for the Effect of Sulpiride on Puerperal Lactation and Maternal Serum Level of Prolactin



Abstract
The effects of oral administration of sulpiride on prolactin secretion and initiation of puerperal lactation were studied. Ten women were given 50 mg orally twice a day for 5 days from 2nd to 7th day of puerperium (Sulpiride group). Ten women were given a placebo in the same way (Control group).
Daily and total amounts of breast milk were measured before and serially during trial. Serum prolactin concentration was measured on immediately after delivery, 1st, 3rd, 5th, and 7th day of puerperium.
The results were as follow;
1) The daily milk yield of control and sulpiride group on 3rd, 4th, 5th, 6th, and 7th day of puerperium were 16.9 ml and 55.0 ml (p<0.01), 70.0 ml and 147.0 ml (p<0.005), 111.4 ml and 222.9 ml (p<0.005), 167.8 ml and 261.4 ml (p<0.005), 163.6 ml and 322.1 ml (p<0.005), respectively.
These results suggested the statistically significant increase of daily milk yield in the sulpiride group in comparison with control group.
2) Mean (¡¾SD) total milk yield for the 3rd to 7th day of puerperium of sulpiride and control group were 1008.6¡¾216.1 ml and 532.6¡¾132.0 ml.
3) Control and sulpiride group were divided into three sub groups of less than 500 ml, 501 to 1000 ml, and more than 10 ml according to total milk yield. The number of patients belonged to more than 1000 nil group were 5 in sulpiride group, but 1 in control group.
4) Mean (¡¾SD) serum prolactin concentration of control and sulpiride group on immediately after delivery. 1st. 3rd, 5th and 7th day of puerperium were 90.98¡¾20.84 ng/ml and 103.67¡¾ 16.89 ng/ml. 105.09¡¾¢¥23.02 ng/ml and 107.43¡¾14.39 ng/ml, 125.84¡¾29.34 ng/ml and 167.80¡¾12.50 ng/ml, 152. 60¡¾16.33 ng/ml and 186.62¡¾16.42 ng/ml, and 146.01¡¾27.06 ng/ml and 186.63¡¾15.40 ng/ml, respectively.
The maternal serum levels of prolactin immediately after delivery and 1st day of puerperium were same in both groups, but that of 3rd, 5th and 7th day of puerperium were significantly increase in the
